Contraindications

Neurological

  • epilepsyrelative

    Active seizure disorder or epilepsy. Serotonergic psychedelics may reduce seizure threshold, though direct evidence is limited. Risk is significantly amplified by concurrent lithium or tramadol use.

Psychiatric

  • bipolar disorderabsolute

    Bipolar I disorder, particularly with history of psychotic features, represents an absolute contraindication. Bipolar II carries elevated risk. Serotonergic psychedelics can destabilize mood cycling and precipitate mania.

  • psychotic disordersabsolute

    Personal or family history of schizophrenia, schizoaffective disorder, or other psychotic spectrum disorders. 5-HT2A agonists can trigger acute psychosis or prolonged psychiatric destabilization in predisposed individuals. This is the most consistently documented psychiatric risk of the serotonergic psychedelic class.

Pregnancy & Breastfeeding

    No reproductive or developmental toxicity data exist for PRO-LAD. Ergoline-class compounds have theoretical uterotonic potential. Contraindicated during pregnancy and breastfeeding as a standard precaution.

Other

  • concurrent lithium useabsolute

    Concurrent lithium use is an absolute contraindication. The LSD + lithium combination is associated with seizures in case reports. This risk extends to PRO-LAD by class analogy as a 5-HT2A agonist lysergamide.

  • concurrent tramadol useabsolute

    Concurrent tramadol use presents dual risk: serotonin syndrome from combined serotonergic load, and compounded seizure threshold reduction. Classified as dangerous interaction.

  • concurrent MAOI useabsolute

    Concurrent use of monoamine oxidase inhibitors (MAOIs) potentiates serotonergic activity and presents serotonin syndrome risk. Both irreversible (phenelzine, tranylcypromine) and reversible (moclobemide) MAOIs are contraindicated.
