Cardiovascular

No DPT-specific cardiovascular data exist. Class-level risk from serotonergic cardiovascular stimulation.
Neurological
- epilepsyabsolute
DPT exhibits dose-dependent proconvulsant effects confirmed in animal models. The fatal case report involved seizure at approximately 2 hours post-insufflation. Any pre-existing seizure disorder substantially increases risk.
Psychiatric
- bipolar disorderabsolute
History of bipolar disorder (type I or II) is an absolute contraindication due to manic episode precipitation risk.

Personal or family history of schizophrenia, schizoaffective disorder, or other primary psychotic disorders is an absolute contraindication.
