Contraindications

Cardiovascular

  • severe cardiovascular diseaserelative

Severe or unstable cardiovascular disease. LSD/ALD-52 produces sympathomimetic stimulation that may pose risk to individuals with pre-existing cardiac conditions, though the cardiovascular effects are modest in healthy populations.

Neurological

  • concurrent lithium useabsolute

Concurrent lithium use. Case reports associate lithium + psychedelic combinations with seizures. The mechanism is not fully characterized but involves adverse interaction between lithium's neuromodulatory effects and psychedelic-induced cortical excitation.

  • epilepsyrelative

Epilepsy or history of seizure disorders. Theoretical risk based on 5-HT2A-mediated cortical excitation. Clinical significance at standard doses is uncertain.

Psychiatric

  • psychotic disordersabsolute

Personal or family history of schizophrenia, schizoaffective disorder, or other primary psychotic disorders. Serotonergic psychedelics can trigger acute psychosis or unmask latent vulnerability.

  • bipolar disorderabsolute

Bipolar disorder, especially with psychotic features. Risk of triggering manic episodes or psychotic decompensation.

Pregnancy & Breastfeeding

  • pregnancyabsolute

Pregnancy. No ALD-52 or LSD teratogenicity data are sufficient to establish safety. Contraindicated by precautionary principle.

Other

  • concurrent tramadol useabsolute

Concurrent use of tramadol. The combination creates risk of serotonin syndrome (clonus, hyperreflexia, hyperthermia, agitation) which is life-threatening in severe cases.

  • concurrent MAOI useabsolute

Concurrent use of monoamine oxidase inhibitors. Dangerous potentiation of serotonergic effects with risk of serotonin syndrome.
