Contraindications

Cardiovascular

  • cardiac valve diseaseabsolute

Pre-existing cardiac valve disease is an absolute contraindication due to 6-APDB's confirmed 5-HT2B agonism, which directly promotes valve fibrosis through TGFβ-dependent mitogenic signaling.

  • pulmonary arterial hypertensionabsolute

Pulmonary arterial hypertension is an absolute contraindication. 5-HT2B activation promotes pulmonary vascular remodeling.

  • uncontrolled hypertensionrelative

Uncontrolled hypertension is a relative contraindication due to norepinephrine-releasing activity producing additional cardiovascular stress.

Neurological

  • epilepsyrelative

Epilepsy or seizure disorders are a relative contraindication. Monoamine release and stimulant effects may lower seizure threshold.

Psychiatric

  • bipolar disorderrelative

Bipolar disorder is a relative contraindication. Stimulant and serotonergic effects may trigger manic or mixed episodes.

  • psychotic disordersrelative

Personal or family history of psychotic disorders is a relative contraindication. Monoamine release may trigger psychotic episodes in predisposed individuals.

Hepatic

  • hepatic impairmentrelative

Hepatic impairment is a relative contraindication. Reduced CYP enzyme activity may result in higher and more prolonged drug exposure.

Pregnancy & Breastfeeding

  • pregnancy and breastfeedingabsolute

Pregnancy and breastfeeding are absolute contraindications. No reproductive safety data exist for 6-APDB; class-level risks from monoamine release apply.
