Severe respiratory disease, COPD with respiratory insufficiency, or conditions requiring supplemental oxygen. Based on animal model evidence; clinical significance at standard human doses not established.

Active epilepsy or history of seizure disorders. Tryptamine compounds can produce convulsions at supratherapeutic exposures.

Personal history of schizophrenia, schizoaffective disorder, or other primary psychotic disorders. Family history represents a relative contraindication. Standard class-level contraindication for all serotonergic psychedelics.

Bipolar disorder type I or II. Clinical trials of psychedelics routinely exclude bipolar disorder. Rapid emotional amplification and ego dissolution may destabilize mood regulation.

Pregnancy and lactation are contraindicated in the absence of any reproductive toxicity or teratogenicity data.

Concurrent or recent use of any monoamine oxidase inhibitor is an absolute contraindication. This includes pharmaceutical MAOIs (phenelzine, tranylcypromine, isocarboxazid), reversible MAO-A inhibitors (moclobemide), and beta-carboline alkaloids (harmaline, harmine in ayahuasca). The pharmacokinetic interaction is quantified and the serotonin toxicity risk is potentially lethal.
