Cardiovascular
- Cardiovascular diseaserelative
ischemic heart disease, uncontrolled hypertension, arrhythmias
LSD produces modest increases in heart rate and blood pressure at recreational doses. These effects are clinically insignificant in healthy individuals but may pose risk in persons with pre-existing cardiovascular disease. No direct cardiovascular data exist for 1P-ETH-LAD or ETH-LAD.
Psychiatric
- Psychotic disordersabsolute
schizophrenia, schizoaffective disorder, bipolar I disorder with psychotic features
Personal or family history of psychotic disorders is an absolute contraindication for all classical serotonergic psychedelics. 5-HT2A receptor agonism can trigger acute psychotic episodes, and individuals with genetic predisposition are at elevated risk. This is the most critical psychiatric contraindication for the psychedelic class.
- Anxiety disordersrelative
generalized anxiety disorder, panic disorder, social anxiety disorder
Anxiety disorders increase the risk of acute psychological distress during the psychedelic experience, particularly panic attacks and psychological destabilization. Set and setting management is critically important but may not fully mitigate risk in anxiety-disordered individuals.
Pregnancy & Breastfeeding

No reproductive toxicity, teratogenicity, or developmental toxicity data exist for 1P-ETH-LAD or ETH-LAD in any species. Standard precautionary principle applies: avoid during pregnancy and breastfeeding.
Other
- Concurrent lithium useabsolute
Concurrent lithium use is an absolute contraindication. The lithium–LSD class combination carries well-documented risk of seizures and dangerous interactions. This applies to all serotonergic psychedelics including 1P-ETH-LAD after prodrug conversion to ETH-LAD.
- Concurrent MAOI useabsolute
Concurrent use of monoamine oxidase inhibitors (MAOIs) is an absolute contraindication due to the risk of serotonin syndrome — a potentially life-threatening condition characterized by hyperthermia, autonomic instability, and neuromuscular hyperactivity. MAOIs potentiate all serotonergic psychedelics by blocking serotonin catabolism.
- Concurrent serotonergic medicationsrelative
SSRIs, SNRIs, tramadol, triptans
Concurrent use of serotonergic medications (SSRIs, SNRIs, tramadol, triptans) presents variable interaction risk. Chronic SSRI use may blunt psychedelic effects through 5-HT2A receptor downregulation. Acute co-administration of any serotonergic agent with 1P-ETH-LAD increases serotonin syndrome risk. Tramadol specifically is classified as dangerous due to its dual serotonergic/opioid mechanism.
