Contraindications

Cardiovascular

  • Cardiovascular diseaserelative

    Serotonergic psychedelics produce mild sympathomimetic cardiovascular effects including tachycardia and transient blood pressure elevation. While clinically insignificant in healthy individuals at standard doses, these effects may be poorly tolerated in individuals with pre-existing cardiovascular conditions. Inferred from LSD class data; cardiovascular effects of 1cP-MiPLA have not been studied.

Psychiatric

  • Personal or family history of psychosis or schizophrenia spectrum disordersabsolute

    Classical serotonergic psychedelics acting via 5-HT2A agonism carry a well-documented risk of precipitating acute psychotic episodes in individuals with personal or family history of schizophrenia spectrum disorders. As a presumed prodrug for MiPLA (a 5-HT2A agonist), 1cP-MiPLA inherits this contraindication by class analogy from LSD.

  • Bipolar disorderabsolute

    Serotonergic psychedelics can destabilize mood regulation in bipolar spectrum conditions, risking precipitation of manic or mixed episodes. This contraindication is inferred from the classical psychedelic class (LSD primary reference) and applies to 1cP-MiPLA as a presumed 5-HT2A agonist prodrug.

Pregnancy & Breastfeeding

  • Pregnancyabsolute

    No reproductive toxicity data exist for 1cP-MiPLA, MiPLA, or LSD. The absence of safety data in pregnancy warrants a precautionary absolute contraindication. The ergoline scaffold has historical associations with uterotonic activity (ergot alkaloids), though LSD itself has not been demonstrated to cause birth defects in controlled studies.

Other

  • Concurrent tramadol useabsolute

    Tramadol is a serotonin-norepinephrine reuptake inhibitor in addition to its opioid activity. Co-administration with serotonergic psychedelics creates risk of serotonin syndrome, characterized by neuromuscular abnormalities, altered mental status, and autonomic hyperactivity. Fatalities from tramadol combined with serotonergic agents have been reported. The dangerous interaction rating is supported by established mechanism, though the specific 1cP-MiPLA + tramadol combination has not been studied.

  • Concurrent MAOI useabsolute

    Monoamine oxidase inhibitors block the enzymatic degradation of serotonin. Combined with a serotonergic psychedelic that directly agonizes 5-HT receptors, this creates a high risk of serotonin syndrome. Fatalities have been reported with MAOI + serotonergic substance combinations. This contraindication applies to all classical serotonergic psychedelics including 1cP-MiPLA by class analogy.

  • Concurrent lithium userelative

    Case reports suggest an increased risk of seizures when lithium is combined with classical psychedelics. The mechanism is not fully established. This is a relative contraindication based on limited case-report-level evidence extrapolated from the LSD class.
